چکیده

When we used plasma containing EDTA as an anticoagulant, the growth of Lacto bacillus casei was inhibited in the folate microbiological assay involving use of 300 p.L of assay mixture in a 96-well microtiter plate (1-3). We postulated that this inhibition was caused by EDTA chelation of manganese, iron, and magnesium ions, all of which are essential for growth of L. casei (4, 5). The singlestrength medium manufactured by Difco Laboratories (Detroit, MI) contains manganese, iron, and magnesium in concentrations of 25, 20, and 450 /LmolIL, respectively. Therefore, we tested the effect of adding either manganese, iron, or magnesium to the medium to reverse the growth inhibition of L. casei caused by EDTA. Figure 1 (top) shows L. casei response curves to (6S)-5HCO-H4PteGlu with various concentrations of EDTA. Inhibition of L. casei growth became obvious at an EDTA concentration of 0.65 mmollL. We considered that 2.6 mmol of EDTA per liter would be the maximum concentration possible if we used 50 tL of plasma per assay from 4.0 mL of whole blood drawn into a tube containing 10.5 mg of disodium EDTA. Addition of manganese or iron to the assay mixture restored the full growth of L. casei in the

Cryoprotected Lactobacillus casei: an approach to standardization of microbiological assay of folic acid in serum.

Folate-depleted, cryoprotected preparations of Lactobacillus casei are stable for at least eight months at -18 degrees C, capable of reproducible growth and suitable as a ready source of inoculum for measurement of folates in physiological fluids. Cryoprotected microorganisms can be a commercially available laboratory reagent, thus simplifying and further standardizing the microbiological assay...

Use of glycerol-cryoprotected Lactobacillus casei for microbiological assay of folic acid.

A simple procedure for preparing glycerol-cryoprotected Lactobacillus casei cultures has been developed. L. casei grown in medium supplemented with low concentrations of folic acid (0.3 micrograms/L) is diluted with an equal volume of glycerol (800 mL/L) and stored at -20 degrees C. Growth response of the glycerol-cryoprotected L. casei to low concentrations of folic acid exceeded that of cult...

Comparison of radioassay and microbiological assay for serum folate, with clinical assessment of discrepant results.

Folate assays by use of radiolabeled folate provide obvious practical advantages over the standard microbiological assay, but remain incompletely tested. We therefore compared results for 415 sera with a kit involving 3H-labeled folate and the Lactobacillus casei microbiological method. We examined the patients' data when there were discrepancies between the two methods. Although the correlatio...

The present study was designed to examine the effects of Lactobacillus plantaraum and L. paracasei on the growth and aflatoxin-producing ability of Aspergillus parasiticus. In these experiments, direct and indirect interactions of two different probiotic lactobacilli with A. parasiticus was examined.A co-culture system which supported both fungal growth and probiotic bacteria showed the inhibit...
